Nitrogen (N) mineralization is a spatially variable and difficult component of the N cycle to quantify accurately under field conditions. Net N-mineralization was compared by direct measurement, indirect estimate, and laboratory incubation for a restored tallgrass prairie and for deficiently and optimally N-fertilized, no-tillage (NT) and chisel-plowed (CP) maize (Zea mays L.) agroecosystems on Plano silt loam soil (fine-silty, mixed, superactive, mesic Typic Argiudoll) in Wisconsin, USA. Four years of in-situ field measurements using an incubated-soil-core/ion-exchange-resin-bag technique showed that land use significantly affected net N-mineralization. Net N-mineralization was consistently smaller in the restored prairie than in the maize agroecosystems and typically larger in the CP than in the NT maize agroecosystems. Three independent methods for indirectly estimating annual net N-mineralization (i.e., N budget residual, deficiently N-fertilized plant N uptake, and profile-scaled in-situ field measurements) were relatively consistent at capturing land-use and tillage effects on net N-mineralization. Laboratory incubation and periodic leaching of Fall-sampled soils demonstrated that both mineralized N and labile C were co-limiting factors influencing N-mineralization in agricultural soils and generally supported field measurements by showing a significant difference in net N-mineralization with and without added fertilizer-N.  相似文献

Although there is a scarcity of supporting empirical evidence,it has long been suspected that calanoid copepods use mechanoreceptionto detect the presence and location of potential prey itemsentrained in the feeding current. In this study, we documentthe first observations showing a freely swimming calanoid copepod,Skistodiaptomus oregonensis, attacking prey-sized, non-motile,inert particles entrained in the feeding current before theparticles contact the copepod's sensory appendages. Feedingcurrent geometry, fluid velocities and associated behavioursthat characterize these interactions are described. The resultsof this study show how copepod swimming behaviour, coupled witha low-velocity feeding current, not only increases copepod encounterrates with inert prey by increasing direct contact rates, butalso increases the probability of detecting and capturing remotelylocated prey that have well-developed escape responses. In turbulentregimes, a far-reaching, low-velocity feeding current shouldincrease encounter rates, but only if coupled with behavioursthat quickly minimize separation distances once prey is detected.  相似文献

Emancipating sense codons toward a minimized genetic code is of significant interest to science and engineering. A key approach toward sense codon emancipation is the targeted in vitro removal of native tRNA. However, challenges remain such as the insufficient depletion of tRNA in lysate‐based in vitro systems and the high cost of the purified components system (PURE). Here we used RNase‐coated superparamagnetic beads to efficiently degrade E. coli endogenous tRNA. The presented method removes >99% of tRNA in cell lysates, while partially preserving cell‐free protein synthesis activity. The resulting tRNA‐depleted lysate is compatible with in vitro‐transcribed synthetic tRNA for the production of peptides and proteins. Background and Objectives

Hookworm infection is among the major causes of anaemia in poor communities, but its importance in causing maternal anaemia is poorly understood, and this has hampered effective lobbying for the inclusion of anthelmintic treatment in maternal health packages. We sought to review existing evidence on the role of hookworm as a risk factor for anaemia among pregnant women. We also estimate the number of hookworm infections in pregnant women in sub-Saharan Africa (SSA).

Methods

Structured searches using MEDLINE and EMBASE as well as manual searches of reference lists were conducted, and unpublished data were obtained by contacting authors. Papers were independently reviewed by two authors, and relevant data were extracted. We compared haemoglobin concentration (Hb) according to intensity of hookworm infection and calculated standardised mean differences and 95% confidence intervals. To estimate the number of pregnant women, we used population surfaces and a spatial model of hookworm prevalence.

Findings

One hundred and five reports were screened and 19 were eligible for inclusion: 13 cross-sectional studies, 2 randomised controlled trials, 2 non-randomised treatment trials and 2 observational studies. Comparing uninfected women and women lightly (1–1,999 eggs/gram [epg]) infected with hookworm, the standardised mean difference (SMD) was −0.24 (95% CI: −0.36 to −0.13). The SMD between women heavily (4000+ epg) infected and those lightly infected was −0.57 (95% CI: −0.87 to −0.26). All identified intervention studies showed a benefit of deworming for maternal or child health, but since a variety of outcomes measures were employed, quantitative evaluation was not possible. We estimate that 37.7 million women of reproductive age in SSA are infected with hookworm in 2005 and that approximately 6.9 million pregnant women are infected.

Conclusions

Evidence indicates that increasing hookworm infection intensity is associated with lower haemoglobin levels in pregnant women in poor countries. There are insufficient data to quantify the benefits of deworming, and further studies are warranted. Given that between a quarter and a third of pregnant women in SSA are infected with hookworm and at risk of preventable hookworm-related anaemia, efforts should be made to increase the coverage of anthelmintic treatment among pregnant women.  相似文献

Here we present a comprehensive method for proteome analysis that integrates both intact protein measurement ("top-down") and proteolytic fragment characterization ("bottom-up") mass spectrometric approaches, capitalizing on the unique capabilities of each method. This integrated approach was applied in a preliminary proteomic analysis of Shewanella oneidensis, a metal-reducing microbe of potential importance to the field of bioremediation. Cellular lysates were examined directly by the "bottom-up" approach as well as fractionated via anion-exchange liquid chromatography for integrated studies. A portion of each fraction was proteolytically digested, with the resulting peptides characterized by on-line liquid chromatography/tandem mass spectrometry. The remaining portion of each fraction containing the intact proteins was examined by high-resolution Fourier transform mass spectrometry. This "top-down" technique provided direct measurement of the molecular masses for the intact proteins and thereby enabled confirmation of post-translational modifications, signal peptides, and gene start sites of proteins detected in the "bottom-up" experiments. A total of 868 proteins from virtually every functional class, including hypotheticals, were identified from this organism.  相似文献

Two soybean varieties (early-maturing group V and late-maturing group VII) and two cotton varieties (conventional and transgenic (Bt) were grown in adjacent replicated large field plots (approximately 0.1 ha each) at two locations for 3 yr. The dynamics and relative abundance of phytophagous stink bugs within these two crops were observed. The most abundant pentatomid species in both crops for all 3 yr were Nezara viridula (L.), Acrosternum hilare (Say), and Euschistus servus (Say). Several other species also were commonly collected. This is the first record of Mormidea lugens (F.) on soybean and E. quadrator Rolston, E. obscurus (Palisot), Holcostethus limbolarius (St?l), and Oebalus pugnax (F.) on cotton. Stink bugs began arriving in soybean when plant growth stages ranged from pod formation to full seed development. Peak numbers of these insects were found in soybean from the time of full-size seeds in the pods until early maturity. The bugs were first attracted to the earlier maturing cultivar (group V), where they remained until plants began to mature (R7). The pentatomids then moved to the later-maturing cultivar (group VII) as it reached full pod to full seed. Stink bugs began arriving in cotton from the time of the earliest flowers until after the first bolls formed. Peak numbers in cotton occurred during the time when all stages of developing bolls were present. Stink bug numbers were much greater in soybean than in cotton over all three seasons. This preference for soybean over cotton indicates the potential use of soybean as a trap crop for attracting stink bugs away from cotton. Additionally, the coordinated use of early- and late-maturing soybean cultivars as a trap crop could minimize the area requiring insecticides, as well as the number of insecticide applications to cotton.  相似文献

The value of a geographical perspective to infectious disease epidemiology and control has long been recognized. However, the labour required to produce maps, and keep them up to date, has inhibited the development of this area, and very little is currently known about the spatial distribution of parasitic infections other than malaria, trypanosomiasis and onchocerciasis. A recent initiative by an international group of collaborators is attempting to redress the absence of detailed spatial information on the major helminth infections of humans. In this article, Simon Brooker and colleagues describe progress made by this initiative in mapping helminth infections in sub-Saharan Africa, highlighting the value as well as the limitations of this empirical mapping approach.  相似文献
